Our goal is to create and validate a UV Spectrophotometric method for estimating the Ondansetron HCL. According to ICH criteria, the procedure was verified. At 248.8nm, ondansetron has its highest absorption. Between 2 and 22g/ml, the linearity was noted. Recovery studies supported the analysis' findings. The findings of the validation parameters showed that the suggested technique was determined to be exact, precise, repeatable, sensitive, and appropriate for regular quality control analysis for estimation of ondansetron hydrochloride in solid dose formulation.

The current work focuses on the development and validation of a UV spectrophotometric technique for determining chlorpropamide in bulk and formulation. According to ICH criteria, UV spectrophotometric method was validated for a number of parameters, including linearity, precision, accuracy, sensitivity, limit of detection (LOD), and limit of quantification (LOQ). The maximum wavelength of chlorpropamide was discovered to be 580 nm, and a colorimetric method was created for the quantitative estimation of colour and to quantify the absorbance of a particular wavelength of light by a particular solution. The maximal wavelength of chlorpropamide in the complex mixture was discovered to be 710 nm. The UV spectrophotometric technique and colorimetry are simple, sensitive, accurate, repeatable, and exact. Chlorpropamide in bulk can be determined effectively using the suggested method.

Development of UV spectrophotometric method for estimation of curcumin in bulk drug and nanogel formulation: A hydrolytic degradation studies

The main purpose of this research was to develop and validate a cost effective UV spectrophotometric method for determination of curcumin in bulk drug and nanogel formulation. The method was validated according to the guideline of International Conference on Harmonisation (ICH). The curcumin was found to be soluble in methanol with λmax of 425 nm. Curcumin over the concentration level of (1-8 µg mL) depicted linear relation in the plot of concentration vs. absorbance. The regression equation of graph was y = 0.1137x - 0.0138 with R value of 0.999. The limits of detection and quantification were 0.23 μg mLand 0.75 μg mL, respectively. The method was accurate, precise, reproducible and robust since all the samples analyzed had relative standard deviation less than 2%. No statistically significant difference between theoretical and measured concentrations was detected. The degradation kinetics of free curcumin (k = 0.042 µg mL.h, 0.53 µg mL.h and 0.068 µg mL.h) and nanogel formulation (k = 0.178 h, 0.003 h and 0.005 h) were zero and first order, respectively. The proposed method was found to be specific while estimating curcumin loaded nanogel formulation without interference of excipients. The developed method was cost effective and can be used for routine quality control analysis of curcumin.

DEVELOPMENT AND VALIDATION OF UV SPECTROPHOTOMETRIC METHOD FOR ESTIMATION OF TRANS-RESVERATROL IN ETHOSOMES

The present work describes a simple, accurate and validated UV spectrophotometric method for determination of trans-resveratrol loaded in ethosomal formulation. The method was validated for different parameters like linearity, precision, specificity, accuracy, limit of detection (LOD), limit of quantitation (LOQ) and robustness as per ICH guidelines. The wavelength maximum of resveratrol in phosphate buffer pH 7.4 was found to be at 307 nm. The method was found to be linear in the range of 8 to 38 µg mL-1 with a correlation coefficient (r2 ) of 0.9957. The accuracy of the method was studied by recovery study and % recovery was found to be in the range of 99.24 to 100.14 %. The method is simple, accurate and requires relatively inexpensive instruments. The proposed method can be successfully used for determination of trans-resveratrol loaded into ethosomes.

A 96-Well Plates-based UV Spectrophotometric Assay for Determination of Total Glucosinolates in Isatis indigotica Roots

Background: Glucosinolates (GLS) are important secondary metabolites in Cruciferae vegetables and herbs. Currently, the assays of total GLS determination are cumbersome (requiring acidic or enzymatic hydrolysis and addition of staining reagents), time-consuming, and indirect. High concentrations of inorganic salts are inevitably incorporated into the GLS products during separation. There is a need for a quantitative method for simple and rapid determination of total GLS after desalting process. Methods: A 96-well plates-based UV spectrophotometric method for determination of total GLS of Isatis indigotica roots was developed in the present study. The detection wavelength is set at 230 nm using quartz plates. This assay was validated using gluconapin and sinigrin as reference standards, and applied to determine the total GLS of I. indigotica roots prepared from five different desalting methods. Results: This assay is specific for total GLS prepared from I. indigotica roots, and it has acceptable accuracy (91.76–98.18% for quality control, and 95.59–102.52% for addition/recovery), precision (0.24–0.70% pooled RSD), reproducibility (0.31–1.84% RSD), and stability (0.24–1.45% RSD) over a 72-h period. Conclusion: The 96-well plates-based UV spectrophotometric assay is simple and accurate for high-throughput determination of total GLS.

Development and Validation of UV Spectrophotometric Method for Determination of Tacrolimus in Bulk and Capsule Dosage Form

Background The present study aimed to develop and validate a rapid robust effective specific and accurate UV-Vis method for the determination of Tacrolimus in pharmaceutical dosage forms according to International Conference on Harmonization ICH Q2 R1 guidelines.Methodology The method development was done using phosphate buffer of pH 6.8. The method validation parameters like linearity precision accuracy robustness and ruggedness were assessed for the developed method. Pure drug solutions were prepared using Phosphate buffer pH 6.8 the aliquots of concentration range 1-10 microgmL and therefore the linear regression coefficient R2 was 0.999.Results The optimum lambdamax was at 295 nm. The developed method was precise within the interday and intraday studies and showed percentage RSD of 1.731 and 1.243 respectively. Thus a precise simple and cost-effective UV-Vis method for the determination of Tacrolimus was developed.Conclusion The method developed was found to be precise robust simple and cost-effective for the determination of Tacrolimus in bulk and in pharmaceutical dosage forms with regression coefficient R2 of 0.999.

Development and Validation of Three Different UV Spectrophotometric Methods for Determination of New Antifungal Combination Used in Treatment of External Fungal Infections and Their Comparison Using ANOVA

Luliconazole and Terbinafine hydrochloride combination is used for external fungal infections and is newly introduced in market. Hence it is necessary to develop a appropriate method of analysis for rapid and accurate determination of these drugs. Three simple, sensitive, accurate, precise and economical UV-spectrophotometric methods (A,B,C) have been developed for simultaneous estimation of Luliconazole and Terbinafine hydrochloride in their pharmaceutical dosage form and their comparison by ANOVA. Method (A) is well based on first order derivative spectrophotometric method, in which zero crossing points for Luliconazole is 249.50 nm and Terbinafine hydrochloride is 258.50 nm. Linearity was found in range of 3-18 µg/ ml for both the drugs using methanol as a common solvent. Method (B) is constructed on the principle of dual wavelength method using absorbance difference at 281 and 306 nm for Luliconazole & 262 and 296 nm for Terbinafine hydrochloride. Method (C) is based on norm of Q-absorbance ratio method where iso-absorptive point was obtained at 250 nm and the λ max selected was of Terbinafine hydrochloride 282 nm. The developed methods were validated as per ICH guideline. Accuracy and precision of methods were determined and validated. All the developed methods showed good reproducibility and recovery with % RSD <2. These three methods developed were compared using one way ANOVA and the fcal value was found to be less than ftab value indicating that there is no significant difference in the assay results of the three methods. All three methods were found to be rapid, specific, precise and accurate and found no interferences from the cream excipients so it can be used for routine investigation of both drugs in quality control laboratories.

Zero-order derivative and area under curve uv-spectrophotometric methods for determination of calcium levofolinate in bulk and pharmaceutical dosage form

Simple, accurate, and precise UV-Spectrophotometric methods were developed for the estimation of Calcium levofolinate (CLF) in bulk and parenteral dosage form by Zero Order (Method A), Area Under Curve (AUC) (Method B). The solution of standard and sample was prepared in 0.1N sodium hydroxide (NaOH) and further dilution was carried out using Double Distilled Water (DDW). The quantitative determination of the drug was carried out by using the zero-order derivative values measured at 282±2nm (method A) and the AUC method values measured at 270.00nm - 297.80nm (method B). Calibration graphs constructed at their respective wavelength of determination were linear in the concentration range of CLF using 3–18μg/mL (r2=0.999) for zero-order and AUC spectrophotometric methods. Moreover, all these methods proposed were thoroughly validated with ICH guidelines. To summarize, developed spectrophotometric methods in this research are simple, precise, accurate, and sensitive to the assay of CLF in injections.

Development and Validation of Three Novel UV Spectrophotometric Methods for Determination of Newly Discovered Combination for the Treatment of Anxiety and Comparison Using Anova

Paroxetine HCl and Etizolam is a newly approved combination which is used for treating anxiety. It is necessary to develop a method to estimate both drugs in combined dosage form. For simultaneous estimation of Paroxetine HCl and Etizolam three simple, precise and sensitive UV spectrophotometric methods were developed in the present work. Method 1 first order derivative methods, ZCP selected were 241.50 nm for Etizolam and 256.50 nm for Paroxetine HCl. Method 2 is absorbance correction method, 243 n and 295 nm were used to estimate the both the drugs. Method 3 is dual wavelength method, 230 nm and 250 nm were selected for estimation of Paroxetine HCl and 221.50 nm and 288 nm were selected for estimation of Etizolam. Calibration curve of Paroxetine HCl and Etizolam had shown linearity over the concentration range of 25-150 µg/ml and 1-6 µg/ml, respectively, in all three methods. All the three methods were validated as per ICH guidelines and all the results were found within limits. By using ANOVA three methods were compared and it was found that F_cal is less than F_tab which indicates that there was no significant difference observed between the assays obtained by all three methods. All the three proposed methods can be used for its intended purpose because they are highly sensitive, accurate and precise.

Chemometric-assisted UV spectrophotometric method for determination of N, N- diethyl-3-methylbenzamide in insect repellents.

In recent years outbreaks of vector-borne diseases have caused great concern to the population, especially those diseases transmitted by mosquitoes. Repellents appear as an affordable alternative for prevention, making it increasingly important to control the quality of these products, since the content of the active ingredients are directly related to the efficiency and the protection time provided by the repellent. This paper proposes an analytical method for determining the DEET (N, N- Diethyl-3-methylbenzamide) content in insect repellents in lotion using UV spectroscopy. For this propose five different strategies of regression were evaluated: (a) Partial Least Squares (PLS) using full-spectrum; (b) interval PLS (iPLS); Multiple Linear Regression (MLR) with variable selection by the (c) Genetic Algorithm (MLR/GA), (d) Successive Projections Algorithm (MLR/SPA) and the (e) Stepwise (MLR/SW). Appropriate predictions were obtained with RMSEP values between 0.88 and 0.93%w w-1. No systematic error was observed and no significant differences were found between the predicted and reference values, according to a paired t-test at 95% confidence level. The results demonstrated the potential of UV spectroscopy associated to multivariate calibration to determine DEET content in repellents as a fast, simple strategy and with a suitable correlation between the values estimated by the model and the reference values.

Stability-indicating chromatographic and UV spectrophotometric methods for determination of colchicine

Colchicine is an alkaloid that used in the relief of acute gout. Three simple, sensitive, selective and rapid methods have been developed for the determination of colchicine in the presence of its alkaline degradation product with good applicability on its pharmaceutical formulation. The first method was based on TLC separation of colchicine and its degradation using chloroform/methanol/conc ammonia (8:2:0.1 v/v/v) as a mobile phase and densitometric detection at 350 nm. The second method was HPLC using kromasil C18 column and a mobile phase of methanol/ammonia (100:1.5 v/v) with UV detection at 246 nm. The third method was spectrophotometry which involved induced dual wavelength method at 245 nm and 261 nm and first derivative of ratio spectra at 302.4 nm. The developed methods were validated according to ICH guidelines and successfully applied for the determination of colchicine in tablets.

Uv Spectrophotometric Methods for Determination of Sofosbuvir in Pure Form and Pharmaceutical Dosage Forms in Presence of Its Alkaline Degradate

Aims: Two spectrophotometric methods were developed and validated for the determination of sofosbuvir in presence of its alkaline degradate.
 Study Design: Ratio difference and ratio derivative methods were assisted for determination of sofosbuvir in presence of its alkaline degradate, laboratory-prepared mixtures and in tablet dosage forms.
 Place and Duration of Study: Pharmaceutical Analytical Chemistry Department, Faculty of Pharmacy (Girls), Al - Azhar University, between December 2019 and January 2020.
 Methodology: Two analytical methods were achieved and validated for the quantitative determination of Sofosbuvir in presence of its alkaline degradate. The first method was ratio difference (RD) method, where the UV absorption spectra of different concentrations of sofosbuvir were divided by the spectrum of a certain concentration (15 µg mL-1) as a devisor of its alkaline degradate to get the ratio difference spectra. Afterwards, the peak amplitudes difference between 253.7 and 243.5 nm were measured. The second method was the ratio derivative (1DR) method, where the first derivative of the ratio spectra (1DR) was obtained and its amplitude was measured at 247 and 268 nm. Good linearity was obtained over the concentration range of 3-15 µg mL-1 for the proposed methods. The proposed procedures were adopted for the selective determination of intact Sofosbuvir in presence of up to 80% of its degradation product. Sofosbuvir was exposed to different conditions as alkaline, acidic and oxidative degradation.
 Results: The proposed methods were developed and validated with good linearity range of 3-15 µg mL-1 for both methods, and also with good accuracy and precision. And the obtained results were statistically compared to those obtained by the reported method.
 Conclusion: Sofosbuvir was successfully determined by the proposed ratio difference and ratio derivative methods in bulk powder, laboratory prepared mixtures and tablet dosage form with good accuracy and precision. The methods were validated according to ICH guidelines. The results obtained were compared with those of the reported method and were found to be in good agreement.

UV spectrophotometric method for determination of Bilastine in bulk and pharmaceutical formulation

Three simple, fast and economic UV-spectrophotometric methods were developed for the estimation of Bilastine in bulk and pharmaceutical formulation. For zero order (D0) λmax was measured at 281.60 nm. For first order method (D1) amplitude was measured at 280.20 nm (maxima) and 283.60 nm (minima). Bilastine obeyed Beer's-Lambert's law in the concentration range of 10–140 μg/ml for both zero order and first order methods. The % RSD value for both intra-day and inter-day precision was less than 2%. The methods were validated as per ICH guidelines for linearity, accuracy, precision and were found to be linear, accurate and precise.

Development and Validation of an UV Spectrophotometric Method for Determination of Thiamphenicol in Dosage Form

A novel UV spectrophotometric method has been developed for the determination of thiamphenicol in a solid dosage form. The spectrophotometric detection was carried out at an absorption maximum wavelength of 224 nm using water as a solvent. The method was validated for specificity, linearity, precision, accuracy, robustness, and limit of detection (LOD) and limit of quantitation (LOQ). The detector response for thiamphenicol was linear in the selected concentration range of 5 to 25 μg/mL with a correlation coefficient of 0.9975. The precision (RSD) among six sample preparations was 0.65 and 1.50 % for analysts 1 and 2, respectively. The average recovery was 99.91 ± 0.65%. The LOD and LOQ were respectively 0.59 and 1.99 μg/mL. The results demonstrated that the excipients in thiamphenicol soft capsules did not interfere with the spectrophotometric determination of the drug. Using the Youden and Steiner approach, the method proved to be robust. The proposed method can be conveniently employed for routine analysis of thiamphenicol in bulk drug and soft capsules.
